Hi! Do I need to book seat reservations if they’re optional? For a regional train, say, from Koln to Dortmund, will all of the seats normally be taken or can I be assured that there will be spaces left?

Usually there will be seats left 
beaware of Rush hour then the trains are quite full :)
Note usually you cant reserve regionaltrains, only fasttrains and Highspeedtrains :)
