Any suggestion on the best/shortest route from Amsterdam to Stuttgart. I was hoping there is a night train, but seems not. Thank you

There isn’t a night train. There’s a night train from Amsterdam, stopping in Karlsruhe (relatively close to Suttgart), but it stops therein the middle of the night at 4 A.M. so not really a good option as you’ll have to wait anyway for a daytime connection to Stuttgart.
Most straightforward option is to just take day trains (ICE) from Amsterdam, changing in Cologne, it will be more comfortable and more sleep ensured.
