I was making seat reservations and it came up as 2- 1st class rail pass-plus, does the plus indicate the space assigned?

Standard and Plus are the names that Eurostar give to their travel classes instead of 2nd and 1st.
If you have a 1st class pass you can choose to reserve in either but for the small difference in cost I would say that Plus is worth it.
Nope. The "Plus" is just part of the ticket/fare category, not your seat assignment. Your coach and seat number will be shown separately on the final ticket.
