One of us will be turning 60 on day 3 or 4 of a 4day Eurail pass.  Would she be able to purchase a senior pass or would she have to purchase the pass based on her age at the first day using the pass?

It's based on the age on the first day of the validity period. You wouldn't be able to activate a senior pass before turning 60.
