I have 4 days of interrail global pass. I want to go to Rome from Leipzig. It is like 18 hours long. If I start my travel at 1pm and get into my last train at 8pm, do i need to activate my second day for the trip at 12 am again? or is it completed with just 1 day?

Only the date when you board the train counts. So if you don't change after midnight, then it's only one travel day. The app will do this automatically.
Do take enough time to connect with the night train. You don't want to miss it.
