I have recently bought a 10-day 2nd class global pass with Interrail, and along my journey I am going to travel with the AVE International from Montpellier to Madrid. So my question is, can I upgrade this leg from 2nd class to 1st class?

No, upgrades aren't available for this train unfortunately. You can consider changing your pass to a first class pass. If you bought it less than a week ago you can still refund it, otherwise you'll need to pay the exchange fee.
