I’m travelling from London and I’m planning my trip, I just want to know if I have to add London as my starting destination as well as my ending destination

There's no requirement whatsoever about where you start or end your trip, or where you start or end a travel day.

Okay but what if I’m in London now and want to travel from here to other European countries, and come back to London. I don’t need to put London as my number 1 and last destination?

I guess you are refering to the trip planner here: https://www.eurail.com/en/tripplanner

Basically it is a useless tool to count travel days. You do not have to use it in any way. Just make a spreadsheet of your trip and count on how many days the pass might be useful (feel free to buy separate regular tickets for short journeys).

Off topic: Eurostar requires a 35€ seat reservation on top of your Interrail pass. There is limited availability for passholders, check Rail Europe for example (add rail pass -> Eurail). In summer trains can sell out far in advance. Luckily there are some alternatives.
